چکیده

Introduction Some 3.5 years ago the study of heredity took a chemical turn when Avery and his colleagues discovered that deoxyribonucleic acid-DNA-is the " transforming principle " that converts bacteria from one genotype to another. Watson and Crick's structural model of DNA then provided the basis for investigating its role as the hereditary material. There followed rapid advances in the biochemistry of DNA replication, gene expression, and regulation of genes. Recently, fresh impetus has been given to the study of genetic mechanisms, particularly in higher organisms, by several methodological developments that have opened a new approach to the analysis of chromosomes: site-specific cleavage of DNA by restriction endonucleases and electrophoretic fractionation of the resulting fragments ; recombination, cloning, and amplification of DNA segments from any source; rapid methods for determining the nucleotide sequence of DNA; site-directed in vitro mutagenesis; synthesis of polydeoxynucleotides of predetermined sequence; and the ability to introduce cloned, functioning genes into prokaryotic or eukaryotic cells. As a result of these developments even chromosomes which are largely inaccessible to classical genetic methods can now be analysed piece by piece in chemical detail. Genes and signals can be altered at pre-selected sites, and the functional effect of such alterations determined. And active, synthetic genes can be constructed in vitro by recombination or by chemical synthesis.

Viable deletion mutants of simian virus 40: selective isolation by means of a restriction endonuclease from Hemophilus parainfluenzae.

Resistance of simian virus 40 (SV40) DNA to cleavage by Hemophilus parainfluenzae II (HpaII) restriction endonuclease has been used as a positive, in vitro selection for mutants lacking the one HpaII endonuclease-cleavage site of wild-type SV40 DNA. Each of 10 viable mutants isolated by this procedure multiplies significantly more slowly than wild-type virus and contains a small deletion (80 to...

Reliable detection of DNA CpG methylation profiles by the isoschizomers MspI/HpaII using oligonucleotide stimulators.

The increasing interest in methylation patterns of mammalian DNA demands a simple and reliable method to clearly differentiate between cytosine (C) and 5-methylcytosine (5-MeC) within a specific DNA region. The simplest and most commonly used approach is the analysis with restriction endonucleases exhibiting different methylation sensitivities, like the MspI/HpaII pair of isoschizomeric enzymes...

Comparison of two viable variants of simian virus 40.

The DNAs of two viable strains of simian virus 40, 776 and 777, have been compared by using restriction endonucleases. Differences between the two strains were detected at five separate points on the simian virus 40 genome. One of these differences, in the region of DNA coding for the major viral coat protein, was confirmed by tryptic peptide analysis of coat proteins from the two strains. Some...

A colinear map relating the simian virus 40 (SV40) DNA segments of six adenovirus-SV40 hybrids to the DNA fragments produced by restriction endonuclease cleavage of SV40 DNA.

The simian virus 40 (SV40) DNA segments present in a series of adenovirus-SV40 hybrids have been mapped with respect to the sites of cleavage of SV40 DNA by restriction endonucleases. Two approaches have been used. First, nucleic acid hybridizations were performed between equimolar quantities of the denatured DNAs of SV40 and each hybrid virus and the radiolabeled transcripts of 11 DNA fragment...

The mapping and ordering of fragments of SV40 DNA produced by restriction endonucleases.

SV40 DNA is cleaved by the Eco RII and Hae restriction endonucleases to give rise to two different sets of 16 fragments each. These fragments have been ordered by analysis of the products of redigestion of one set of fragments with another restriction enzyme. The cleavage sites have been precisely mapped based on length measurements of the products of each cleavage. This provides a convenient g...
